预处理和培养温度对陆稻游离小孢子愈伤组织诱导及分化的影响

摘 要:

以陆稻品种‘云2号’和‘云3号’为材料, 分别研究了花药预处理、游离小孢子预处理和小孢子初期培养温度对小孢子愈伤组织产量及绿苗形成数的影响。结果表明, 将游离前的小孢子花药在32 ℃下预处理2 d, 或将游离小孢子在32 ℃下预处理2 d, 均可以提高小孢子的愈伤组织产量; 将游离小孢子置于32 ℃培养2 d后转入25 ℃培养优于只置于25 ℃培养, 可以明显提高小孢子愈伤组织产量及绿苗形成数量。

Abstract:

Two cultivars of upland rice were used to investigate the effects of pretreatments of anther or isolated microspores and the initial culture temperature on callus yield and number of green plantlets. The results showed that 2 d of 32 ℃ pretreatment on anthers or isolated microspores could improve the callus yield. Comparing to continuous culture under 25 ℃, 2 d initial culture under 32 ℃ prior to subculture under 25 ℃ could remarkably improve the yields of the callus and green plantlet.
